Support for the idea that physiological gradients of substances into the tissue may be the operative factors promoting organ initiation in vitro is presented. Evidence for this concept was obtained through measurement of the starch content and respiratory activity of upper and lower segments of tobacco (Nicotiana tabacum L. cv. Wisconsin 38) callus and inversion of the tissue during culture.
