ADP-glucose : α-1,4-glucan α-4-glucosyltransferase was obtained from a marine red alga Serraticardia maxima in a form bound with floridean starch granules. The enzyme catalyzed the transfer of glucosyl residue from ADP-glucose, UDP-glucose and GDP-glucose to floridean starch added as a primer. ADP-glucose was the most efficient glucosyl donor in the reaction. Maltose was produced by β-amylolysis of the glucan synthesized by the algal enzyme. The optimum pH for enzyme activity was at 8.4. The enzyme was not obtained in a soluble form from either the chloroplast extract or the whole algal cell extract. Electron micrographs of algal cells revealed that floridean starch granules are localized exclusively outside chloroplasts. Hence, it appears that most of the synthetase is present outside chloroplasts.
