Examinations were made on the colorimetric determination of liquid and ointment for external use containing naphthiomate-T, an anti-trichophyton agent, and the following procedure was established. To a test tube graduated to 10 ml, 0.2 ml of the liquid or ca. 10 mg of the ointment accurately weighed on a microspatula (both corresponding to less than 200 μg of naphthiomate-T) is placed, 0.5 ml of propanol in which metallic sodium has been dissolved to 2% concentration is added, and the test tube is warmed on a water bath to effect solution. The test tube is attached with an air cooling tube and the heated in a boiling water bath to effect exchange decomposition to form 2-naphthol. When cooled, 0.5 ml of methanol solution containing 0.55N hydrochloric acid is added, followed by 0.7 ml of diazonium test solution (containing 0.2% of sulfanilic acid and 0.25% of sulfamic acid), and the mixture is allowed to stand for 5 minutes by which the solution colors by the formation of orange II. This mixture is diluted to 10 ml with methyl ethyl ketone containing 0.06N hydrochloric acid-formaldehyde and optical density of this solution at pH 1-2 is read at 485 mμ. The content of naphthiomate-T is calculated from the calibration curve prepared beforehand. This method enables determination of 25-200 μg/10 ml of naphthiomate-T and the time required for the whole procedure is approximately 2 hours.
