Simultaneous determination of trapidil and its metabolites, 5-methyl-7-ethylamino-s-triazolo[1,5-α]pyrimidine (TP-1) and 5-methyl-7-amino-s-triazolo[1,5-α]pyrimidine (TP-2) in the serum by high-performance liquid chromatography was described. 5-Methyl-7-dimethylamino-s-triazolo[1,5-α]pyrimidine (IS) as an internal standard, Radial Pak CN as a column and acetonitrile-water-phosphoric acid-triethylamine (320 : 80 : 0.3 : 0.1) as a mobile phase were used. The capacity factors for TP-2,TP-1,IS and trapidil were 3.8,4.4,5.1 and 6.9,respectively. The recoveries of trapidil, TP-1 and TP-2 from the serum were in a range of 95.2-100.8% and the detection limits were 0.02μg/ml for all these compounds. Serum levels of trapidil and its metabolites were determined in human subjects and dogs after oral administration of trapidil. As a result, trapidil, TP-1 and TP-2 were detected in the serum of both human subjects and dogs. Elimination of these compounds from the serum in human subjects was found to be slower as compared with that in dogs.
