The present work was undertaken to clarify the correlation between the tyrosinase activity and ascorbic acid in the body fluid of Lepidopterous insects. 1. The melanosis of the in vitro body fluid was suppressed by addition of ascorbic acid. 2. The rate of oxidation of ascorbic acid was paralleled with the tyrosinase activity of the body fluid. That is, ascorbic acid was rapidly oxidized in the body fluid of the prepupae of Philosamia cynthia ricini and the larvae of Bombyx mori, where the tyrosinase activity was high, but it was slowly oxidized in the body fluid of the larvae of Philosamia and the prepupae of Bombyx, where the tyrosinase activity was low. 3. The oxidation of ascorbic acid was strongly inhibited by phenylthiocarbamide at a low concentration, but not by sodium azide even at a considerably high concentration. On the basis of these data, it will be considered that ascordic acid is oxidized by some substance which contains copper in its chemical configuration, especially by tyrosinase. 4. Considering the results that tyrosinase itself did not oxidize ascorbic acid and that anilin strongly inhibited the oxidation of ascorbic acid, the oxidation of ascorbic acid appears to be due to o-quinone which is produced by the action of tyrosinase. 5. From the fact that o-diphenol was found in the body fluid of prepupae and pupae of Philosamia but was never converted into melanin in vivo, it is assumed that ascorbic acid plays an important role in reducing o-quinone into o-diphenol in vivo. This may be a reason for the failure of the melanosis in vivo.
