The present paper deals with the results of electron-microscopical study of the fine structure of the cortex and the pharynx of Parmecium carried out by means of ultra-thin section. The materials used were Paramecium caudatum and P. aurelia cultured in wheat and soya bean powder infusion plus juice of mashed clover leaves. The preparation procedure was as follows : Living animals were gathered by low power centrifugation, after which the supernatant fluid was quickly replaced with a solution of fixing reagent. In this case 1 per cent O_sO_4 solution adjusted with veronal buffer to pH 7.4 was used. Fixation was performed over periods from 30 minutes to 1 hour at approximately 10℃. The fixed animals were washed rapidly with distilled water and dehydrated by passing through alcohol series and then imbedded in n-buthyl methacrylate catalyzed by 2% benzoyl peroxide. In order to concentrate the animals, low centrifugal forces were used throughout dehydration and imbedding procedure. The materials in this plastic substance were cut into thin sections of about 0.1μ in thickness with a Spencer ultramicrotome. The specimens were examined under a J.E.M. Type III electron microscope made by Japan Electron Optic Laboratory. The results observed are as follows : 1) The cortex is covered externally by a thin sheath (outer-limiting membrane), constituting the pellicle. Beneath the membrane, there is a space, and the periphery of the plasma is limited by a ridgework forming a pattern of polygons with depressed centers. 2) The cilium projects from the depressed center of each polygon. The proximal end of the cilium is connected with a besal body and is inserted vertically into the cortex. Around the base of each cilium there is a transverse plate or a ciliary ring. The ciliary ring seems to be made up two closely apposed concentric circle walls, and the space between the two walls is divided into about nine parts by septa. 3) In the middle of each polygon, an opening of a trichocyst is located. 4) The pharyngeal penniculus consists of eight parallel rows of cilia. The cilia in this area are similar in structure as those in cortex.
