The author (1952) reported that at the early developmental stages of Rhacophorus schlegelii var. arborea the peri-vitelline fluid contains Na-ion, Cl-ion- SO_4-ion, protein, uric acid, and urea. The present study failed to show any trace of creatinide, but revealed the existence of Fe only at the stage before hatching. Protein in the fluid was positive for Biuret reaction, Ninhydrin reaction, and Xanthoproteic reaction, but negative for Hopkins-Cole reaction and Liebermann reaction. Moreover the test for sulfur was positive for the fluid at the stage of neurula and before hatching. Therefore the amino acid, containing S was present in the fluid at these two stages. Total volume of protein precipitated by half saturated (NH_4)_2SO_4, and hydrolysed by 8N-HCl was studied by means of paper chromatography, and the seven spots of amino acids were recognized. They were found to correspond to cystine, tyrosine, lysine, glutamic acid, arginine, aspartic acid, and leucine or isoleucine by means of the color reaction as well as by the comparison with the amino acids prepared by Ajinomoto Manufacturing Co.
