1. The puparium pigmentation of the mutant "yellow" of Drosophila virilis is affected by the yeast content of the culture medium. The greater the content of yeast, the darker becomes the puparium. 2. When cultures were raised on a standard cornmeal-molasses-agar medium containing a minute quantity of yeast (0.05g per 10cc of culture medium), the pupae were light yellow in clour. The addiition of tyrosine or phenylalanine to the medium exerted a marked influence on the puparium. The addition on dopa scarcely affected the pigmentation. Tryptophane and other amino acids also showed no effect. 3. The body weight of the pupae became heavier with an increase in the yeast content, but it was not affected by the addition of tyrosine to the medium. Hence it may be concluded that tyrosine and phenylalanine are used as the precursor of the puparium pigment.
