The author examined the specific affinity of the intracellular substances to Victoria blue 4R. (1) Observations in vitro showed that Victoria blue stained fatty acids and lecithines well, but neither neutral fats nor mineral oils. (2) By the preliminary treatments with depolymerizers of nucleic acids, the staining reaction of nuclei to Victoria blue was always decreased. It was even entirely abolished by the treatment of long duration. This shows that this dye stains the nucleic acids. (3) Victoria blue stained very intensely the granules of basophilic granulocytes. This property of basophilic granules also disappeared following the preliminary treatments with depolymerizing agents for nucleic acids, polysaccharides, and proteins respectively. (4) Even after an extraction for a long period (one week) with alcohol, acetone or ether, the staining intensity of the nuclei and basophilic granules with Victoria blue did not decrease. From the facts above mentioned, it seems unlikely that Victoria blue 4R stains lipids only. By the use of this dye for the lipid staining, the consideration for group reaction is necessary.
