In order to understand the singnificance of the rapid growth of the epithelium through multiplication of cells during the period of the pearl-sac formation, the distribution of alkaline phosphatase was investigated in the mantle and pearl-sac of the fresh water pearl mussel (Hyriopsis schlegelii), with special regard to the mechanism of the calcium and protein secretion from the pearl sac. The mantle edge is usually provided with three tentacle like folds, which are usually designated as the shell hold, the middle fold and the inner fold. Of these folds, the inner fold only was positive in reaction for the alkaline phosphatase test. Namely, the alkaline phosphatase reaction was shown as a brownish black thin layer covering the epithelium. The nuclei of the epithelial cells are also proved positive to the reaction. However, the connective tissues and muscular bandles occurring in the mantle reacted weakly in this test. A part of the mantle piece which is to be used for the implantation in the culture method showed a weak reaction. In the case of the pearl-sac, the connective tissues locating in the inner side of the pearl-sac epithelium and those surrounding the nucleic piece under the pearl-sac was strongly positive for alkaline phosphatase reaction. Particulary, the connective tissues developing around the wound caused by the operation showed a strong reacton. The pearl-sac of the epithelium reacted weakly for this test.
