The general characteristics of a new ascites tumor of rats. the MTK-sarcoma III, were investigated here with special regard to its cytolgical features. 1) The rate of transplantation was found to be 84.4% on an average, based on 77 transplant generations. The average life span of the tumor bearing rats was 13 days. 2) The rate of mitosis in a transplant generation was observed through the whole life span of a tumor rat (Table 1, Chart 1). 3) Generally, the number of dividing cells, particularly those showing well-balanced subdiploid chromosomes, increases during the early dart of a transplant generation, and the mitotic rate shows the highest frequency during the middle part. Then it decreased gradually toward the latter part of the life span. 4) The chromosome numbers of the MTK-sarcoma III show a wide variation ranging from 26 to approximately 82. The majority of cells were found to possess 38-42 chromosomes, among which those showing 40 chromosomes appeared at the highest rate. 5) It was found that there are two strains of tumor stem-cells which are primarily responsible for the formation of the tumor in transplantation. These cells were characterized by having ±40 chromosomes which multiply with regular mitotic behavior. The chromosome complexes characteristic of these two strains of tumor stem cells formulated as thus: i) (26-30) R's + (10-14) V's & J's ii) (27-31) R's + (9-13) V's & J's The cells of the first type are characterized by showing a prominent V-shaped chromosome of the largest size, while the latter contains no such element at all. These, the chromosome constitutions of this tomor differ from those of the Yoshida sarcoma, MTK-sarcoma I and II, but strikingly particular to this tumor. Based on the results of the above observations, it is evident that general characteristics of the MTK-sarcoma III show a striking similarity to those established in the Yoshida sarcoma, MTK-sarcoma I and II in many points.
