The occurrence of karyorrhexis has been observed in the piece of skin epidermis of an adult frog, Rana nigromaculata, immersed in the Ringer's solution containing acetone or alcohol (pH 8). The nuclei in the epidermis showed varied morphological changes according to the layers in which they are situated. A greater amount of the dehydrating agent was found to be necessary for the formation of karyorrhexis in the deeply situated nuclei, than in those of surface layers. The necessary concentration is probably proportional to the degree of hydration of a nucleus, and the karyorrhexis may be taken as a kind of coacervation of nucleoprotein. It is to be expected therefore that the nuclei of a dried frog may show a decrease in the degree of hydration responding quite differently to the action of the dehydrating agents. But the result of the present experiment was as follows: the adult frogs, dried to death by exposure to the air for one day or two, gave up water and lost some 40 per cent of their body weight. Nevertheless, the nuclei in such an epidermis showed exactly the same karyorrhectic reaction as those in the normal frogs with regard to the necessary and optimal concentrations and the degenerative morphology of nuclei, when treated with acetone or alcohol. Therefore, it seems that the bound water contents of the nuclei undergo little change, if the frogs are kept dry.
