Alkaline phosphatase of rat testis fixed with cold acetone was studied, with two histochemical methods; Dempsey's modification of Gomori method, and the azo-dye method derived by Burstone. No significant difference was found in the results obtained by these two techniques, so far as usual procedures were followed. Preparations treated with EDTA (pH 9.2) before incubation, however, showed considerable difference depending upon the technique employed. With the Gomori method, the enzyme activity was hardly detectable unless zinc salt was added to the incubation mixture, whereas the Burstone method revealed positive reaction, probably caused by Zn release from Fast Blue RR salt. These observations suggest that alkaline phosphatase of the testis requires Zn to show its activity. High concentrations of Zn, however, had an inhibitry effect on the enzyme. Cadmium also decreased the activity in the basement membrane, but not in the blood vessels. This may be one of the causes of testes necrosis by Cd administration.
