The activities of glucose-6-phosphatase (G-6-Pase) and of phosphorylase and glycogen content were histochemically examined in the liver of the fetal, newborn, young and adult rats. For the demonstration of phosphorylase and glycogen, the freeze-substitution technique of Chang and Hori (1961, 1962) was used. Although G-6-Pase activity was not demonstrated in the liver of fetal rats, the enzyme activity was detected in the liver of newborn young and adult rats. However, theactivity was progressively decreased with age of the rat. In both newborn and adult rats, the activity was higher in the peripheral area than in the central area of the hepatic lobules. The glycogen content and the phosphorylase activity were higher in the liver of fetal rats than in those of the new born and adults. The liver of both young and adult rats showed higher phosphorylase activity in the peripheral area than in the central area of the lobules (Figs. 4 and 5), whereas no such feature was observed in the liver of fetal and newborn rats. The histochemical method used in the present study may serve as a criterion for understanding chemical changes during the course of cell growth and differentiation.
