For analyzing the responses of retinal neurons on the basis of cell topography, marking of the position of the electrode tip in morphologically identified neurons was achieved by intracellular injection of a mixed solution of Procion Yellow and Water Blue dyes from a single-barrel microelectrode. In fluorescence microscopy, a dark spot of non-fluorescent, less diffusible Water Blue dye indicated a recording site in Procionstained neurons. A similar marking was also obtained by means of a double-barrel microelectrode, one barrel of which was filled with Procion Yellow and the other with a solution of metal ions. In bipolar cells, the electrodes penetrated either the cell soma or the axon terminal, and it was suggested that a synaptic potential arising in the dendrite propagated electrotonically to the terminal. In amacrine cells, the response obtained from the thick dendrite was identical to that of the cell soma, indicating that both parts of the cell were equipotential. Thus, this technique may be useful for a better understanding of neuronal functions in a wide variety of neural preparations.
