The substrate specificities of glucose 6-phosphate dehydrogenase (G6PD) from 86 molluscan species were examined by histochemical stainings of polyacrylamide gels after electrophoresis in order to ascertain whether hexose 6-phosphate dehydrogenase (H6PD) is present in mollusks. Nine species showed no G6PD activity. Of the remaining 77, 35 species possessed G6PD which is active only on nicotinamide adenine dinucleotide phosphate (NADP)+glucose 6-phosphate (G6P). Thirty-one possessed G6PD which is also active on galactose 6-phosphate (Gal6P)+NADP, 4 possessed G6PD active on G6P+NAD, and 7 possessed G6PD active on Gal6P+NAD in addition to the other substrates. The majority of G6PD having broad substrate specificity were found in the Suborders Heteropoda and Gymnoglossa of Order Mesogastropoda. Quantitative analyses of the substrate specificity of the enzymes from seven species indicated that none of the molluscan G6PD resembled H6PD in this respect. We conclude that H6PD does not exist in the Phylum Mollusca. This is consistent with our previous speculation that the divergence of H6PD occurred at the time of echinoderm evolution.
