Ovarioles taken from pupae of the silkworm, Bombyx mori, (variety: N131) 5 days after pupation were cultured either in Grace's insect tissue culture medium (G.I.T.C.M.) or in the same medium containing either ecdysterone or pupal hemolymph. The in vitro formation of the egg shell was compared with the in vivo formation. In the ovarioles of 5-day-old pupae, that is before the commencement of the culture, the formation of the egg shell had not yet started. After 6 days' culture, oocytes had migrated from the ovarioles into the medium, and each oocyte had already been covered with an egg shell. The stratified structure of outer, middle and inner layers and a regional difference in the surface structure of the in vitro egg shell were observed, as in the normal eggs. However, the egg shell produced in vitro was thinner than the in vivo egg shell, especially in the outer layer. The addition of ecdysterone or hemolymph of 5-day-old pupae to the medium had no specific effect on the formation and the structure of the egg shell.
