An Improved Cobalt Iontophoresis Technique for Neuron Tracing

The cobalt iontophoresis technique for neuron tracing was applied to the in vivo optic nerve of the crucian carp. Ammonium sulfide, which is widely used to produce a cobalt precipitate as black sulfide within the neuron, greatly damages the nervous structure of the fish. To overcome this problem we employed H_2S-alcohol solution instead of ammonium sulfide and were able to obtain sections of the nervous tissue without any deformation under light microscopic observation.
