Differentiation of newt somites was studied by the fluorescent antibody method and electron microscopy to learn in detail the interrelationship between the appearances of muscle specific proteins and myofilaments during muscle differentiation. The cells in the somite were all mononucleated until St. 26. Using electron microscopy, filamentous material was observed in the vicinity of the cell membrane at St. 24, and bundles of myofilaments were observed at St. 26. Thereafter, Z-bands (St. 28) and A-, I-bands (St. 33) were observed, and finally M-lines (St. 35) were seen. Figures of cell fusion were observed at St. 28. By the fluorescent antibody-indirect method using the antiserum prepared against chicken skeletal muscle tropomyosin, fluorescence was first detected in the embryonic myotome region of the frontal somite at St. 25, and fluorescent cross striations at St. 31. In every stage, correspondence was observed between the distributions of fluorescence and myofilaments or myofibrils. These facts indicate that tropomyosin and myofilaments appear at almost the same stage.
