In an attempt to establish the cytological basis for the production and secrretion of the frog hatching enzyme, electron microscopic obserbations were made on hatching gland cells (HGCs) in Rana chensinensis embryo. Three types of epidermal cells were identified in the tailbud embryo. Among them HGCs were characterized with prominent microvilli, a number of membrane-limited secretory granules, well developed Golgi complexes and rough endoplasmic reticulum. The localization of HGCs was confined mostly to the frontal region and to a lesser extent along the dorsal mid-line. The cells made its first appearance at stage 17, continued to secrete the granules during stages 18-21, and underwent gradual degradation at the post-hatching stages. Cytochemical tests proved that the polysaccharide-containing granules are produced in the Golge complex during the pre-hatching stages. At the post-hatching stages, however, an acid phosphatase activity became detectable in the granules found in the Golgi complex of HGC. In addition, evidences were presented to show that these (primary) lysosomes fuse with large, membrane-bounded bodies to form phagolysosomes which participate in the degradation of HGCs at the post-hatching stages. These observations provide a good ultrastructural basis for understanding the biochemical aspects of the frog hatching enzyme which were elucidated in recent years.
