Spontaneous diphasic action potentials were recorded by intracellular microelectrode from Opalina collected from autumn and winter frogs, Rana japonica, soon after transference of the cells into the adaptation medium from the frog rectum. These action potentials changed into periodically hyperpolarizing action potentials about one hour after immersion of Opalina in the medium. Repeated diphasic action potentials were also evoked by application of 8mM BaCl_2 (dissolved in the adaptation medium) to the cell in which no active change in the membrane potential was observed. In this case, the inside negativity of the cell decreased as much as about 10mV by application of the BaCl_2 solution (Fig. 1, D). When the BaCl_2 solution was applied to the cell in which the spontaneous hyperpolarizing action potential was produced, the membrane was further hyperpolarized once, then the inside negativity decreased, and repeated diphasic action potentials appeared spontaneously (Fig. 1, C). In spring (May), no diphasic action potentials was recorded but only repeatedly hyperpolarized action potentials were recorded even if the microelectrode was inserted into Opalina immediately after transference from the frog rectum to the adaptation medium, but the hyperpolarized action potentials were changed into the diphasic ones by application of the BaCl_2 solution.
