細胞性粘菌の発生異常突然変異体による分化の研究

By nitrosoguanidine treatment a mutant strain was isolated, which can not differintiate spores under ordinary culture conditions. This strain, designated as KS17, forms fruiting bodies with undifferentiated cells instead of ordinary spores, while the stalks are well differentiated. KS17, however, can form well differentiated spores under a certein experimental condition. It was first observed that when the cell density at the stage of aggregation was very low the cells formed fruiting bodies with normal spores. This seems to suggest that the cells are producing a substance to repress the differentiation of sporess and that the substance is effective when it is accumulated to a certain concentration. To test this possibility, the mutant cells were sown on a millipore filter placed on a wet paper pad in the density of A) 2.8×10^5/cm^2, B) 7.0×10^6/cm^2, C) 4.2×10^7/cm^2. In A) the cells developed into fruiting bodies with normal spores, in B) into fruiting bodies with no differentiation of spores and in C) the cells stopped developement at the slug stage. Removing the cells with the millipore filter, fresh cells were sown on a new millipore filter placed on the old paper pad in a low density of 2.8×10^5/cm^2. The result was same as before: no spore were formed in B) and C) inspite of the low density of the cells. Wild cells were also proved to produce this repressing substance. When wild type cells are sown in a density of higher than 7.0×10^6/cm^2, the substance which repress spore formation in KS17 is accumulated in the pad. Wild type cells are resistant to this substance. Wild type cells sown on the millipore filter on which high density of the cells had been cultured previously can differentiate spore normally. Thus it is concluded that every cells of this species produces the repressing substance and that mutant KS17 is especially sensitive to this substance while the wild type is resistant to it.
