Effects of Nervenruh forte, a psychotropic drug, on mouse embryos from dd strain were investigated with special regard to the frequency of fetal malformation and death. On the 9th day of gestation, pregnant females received Nervenruh forte, by a single oral administration or by a single intraperitoneal injection, at dose-levels of 10mg/kg, 100mg/kg, 200mg/kg and 400mg/kg of body weight. Oral administration-experiments resulted in that teratogenic, embryocidal and growth-inhibiting effects were not significantly different between the experimental and control series. On the other hand, fetuses with external malformations characterized by exencephaly, open eye and exophthalmos appeared in mice received intraperitoneal injection, though the frequency was low. Fetuses with skeletal anomalies represented by rib defects appeared in treated animals only. The difference in frequency of those anomalies between control and intraperitoneal-injection-groups was statisically significant. The mortality of fetuses in intraperitoneal experiments was not different between the control and treated mice, so far as the doses, 10mg/kg and 200mg/kg, are concerned, while the incidence of fetal death at doses of 100mg/kg and 400mg/kg was significantly different between the two groups. No growth-inhibiting effects on the fetuses were obtained at all dose levels. In the scope of the present experiments, a possible conclusion is that teratogenic and embryocidal effects in intraperitoneal experiments may be due to direct action of this drug.
