1. By use of thin layer chromatography, more than 12 amino acids were identified in a hydrolyzate of the chorion. 2. When a piece of chorion was treated with pepsin (5mg/ml), the inner walls of the chorion were digested at regular spacings in an hour, and the chorion rolled up inward. After three hours, fine hexagonal structures on the chorion surface gegun to disapper. Although the outmost layer of the chorion was resistant to pepsin, most of the villi on the chorion were digested within 24 hous. By peptic digestion for 5 hours, large protein fragments were obtained from the chorion. When the egg was fertilized in the Yamamoto's saline solution containing an antiserum against these fragments, more than six layers were recognized in the chorion. 3. To prepare hatching enzyme extract, hatching glands isolated from 200 embryos were cytolyzed in 1 ml distilled water. In the extract, a piece of chorion was digested quickly from both inside and outside, and most of the fine hexagonal structures became extinct within 30 minutes. The outmost layer of the chorion remained insoluble even in this extract. This insoluble layer was easily destroyed by a mechanical procedure. 4. By use of agar-gel electrophoresis, an attempt to fractionate the hatching enzyme was made. After electrophoresis, agar-gel was cut into strips of 5 mm wide, and each of them was incubated with chorion in borate buffer at pH 8.2 for 24 hours. An active fraction was localized in a certain strip, which digested inter-layer cementing substance of the chorion which was then visualized in concentric arrangements of 10 layers.
