Histochemical Localization of Some Enzymes in the Midgut Gland of the Japanese Horseshoe Crab, Tachypleus tridentatus

The histochemical localization of non-specific esterase and acid and alkaline phosphatases was studied on the midgut gland of the Japanese horseshoe crab, Tachypleus tridentatus. Each tubule of the gland consists of simple columnar epithelium which is composed of vacuolate and slender granule-rich cells. The distribution of nonspecific esterase activity corresponded to the granules in slender epithelial cells and alkaline phosphatase activity was localized in disintegrating epithelial cells and in the free and outer surfaces of these epithelia. As to the localization of acid phosphatase activity, the sections were colored evenly throughout the samples, and no clear-cut result was obtained.
