Primary tissue culture of the land slug, Incilaria bilineata was maintained for 2 or 3 months in a medium based on the concentrations of Na, K, Ca and free amino acids in slug hemolymph. No cell division was observed, but the cells survived. Thin medium layer culture which contained 1.0 ml of medium per 5 cm plastic Petri-dish was used. The small minced tissue pieces were explanted per dish. The medium was renewed every third day. This method enabled long term cultivation. The concentration of alanine in the medium increased during 4 days of cultivation to ca. 20 fold that of the incubated control level without cells. When slug hemolymph was added into the medium, it inhibited the migration of slug cells. However, the hemolymph heated at 56℃ for 30 min neither inhibited the migration nor induced cell division.
