Histological, histochemical and autoradiographic studies were carried out to determine the development time of cell types in the pituitary gland of the medaka (Oryzias latipes) embryo and fry. The relationship between the appearance of thyrotrophs in the pituitary and the functional activity of thyroid follicles was investigated in the embryo at different developmental stages and the stage at which initiation of thyroid hormone synthesis took place was determined. When the embryos were kept at 24±1℃, pituitary could not be found at Stage 26 (3 days after fertilization ; by Matsui, 1949). At Stage 29 (6 days after fertilization), the pituitary could be identified. It was closely attached to the diencephalon and contained some neurosecretory materials. The cell types of the pituitary, however, were not distinguishable at this stage. Thyrotrophs and gonadotrophs could be distinguished at 1 and 3 days after hatching, respectively by Gomori's aldehyde fuchsin (AF) reaction. The thyroid follicles were distinguishable at Stage 32 (9 days after fertilization). The uptake of Na^<125>I by the embryo began to increase at Stage 31 (8 days after fertilization) and reached a high level 10 days after hatching.
