Bioconversion of 17_α-Hydroxyprogesterone into 17_α, 20α-Dihydroxy-4-pregnen-3-one and 17_α, 20β-Dihydroxy-4-pregnen-3-one by Flounder (Platichthys flesus) Spermatozoa

Flounder (Platichthys flesus) spermatozoa were incubated with 17_α-hydroxyprogesterone at 20℃ for 18 hr. After extraction of the steroids from the incubation medium, steroid metabolites were analyzed by gas chromatographymass spectrometry (GC-MS). Selected ion monitoring with 25 possible steroid metabolites as standards represented that main metabolites were 17_α, 20_α-dihydroxy-4-pregnen-3-one (17_α, 20_α-P) and 17_α, 20β-dihydroxy-4-pregnen-3-one (17_α, 20β- P). In addition small amounts of androstenedione and 17_α-hydroxy-5β-pregnane-3, 20-dione were detected. Major two metabolites were further identified using full mass spectrograms to be 17_α, 20_α-P and 17_α, 20β-P, respectively. These results indicate that flounder sperm cells show both 20_α- and 20β-hydroxysteroid dehydrogenase activities.
