In the supernatant fraction of the adult Drosophila, phenoloxidase activity was detected by the use of SDS-PAGE and spectrophotometry. The level of phenoloxidase activity increased within six minutes after the initiation of copulation. The length of time of reaction mass formation corresponded to the increase of phenoloxidase activity after the initiation of copulation in a copulated female. The reaction mass formation in mated female Drosophila was, in part, considered to be in relation to phenoloxidase activation after copulation.
