Mature unfertilized eggs (primary oocytes arrested at metaphase of the first meiotic division) of Athalia rosae can be explanted from ovaries and placed on a filter paper soaked with distilled water, or submerged in distilled water where they are activated and start and complete embryonic development. Eggs placed and kept in a saline solution (0.15 M NaCl) are not activated, show no signs of development and deteriorate after a few hours. However once activated in distilled water for 20min, eggs can then be placed in saline solution and development continues. This 20-min period corresponds to the time required by the arrested metaphase nucleus to proceed to telophase. Various ions as well as osmolality are involved in this egg activation process in varying capacities. Other treatments such as brief desiccation, passage through narrow capillary tubing, pricking with a glass needle or lowering pH of the solution are also found to be effective in activating the eggs to various degrees.
