Histochemical techniques, in particular immunohistochemistry and in situ hybridization, have been valuable tools in expanding our knowledge in neurobiology. The high sensitivity and excellent resolution of these methods have permitted detailed analyses of the complex functional organization of the nervous system with its many different cell types and processes. In the present brief summary we would like to focus on some peptides which normally are produced at very low levels or at levels which can not be detected with our histochemical techniques. However, after experimental manipulations, such as mechanical damage or drug treatment, there is a marked upregulation to detectable levels, both of peptides and their mRNAs. We will describe four systems, primary sensory neurons, striatal neurons in basal ganglia, hypothalamic neurons in the paraventricular nucleus, and chromaffin cells in the adrenal gland. (Primary Sensory neurons) It was early noted that transection of the sciatic nerve causes a marked downregulation of substance P levels in the corresponding dorsal root ganglia. Subsequently numerous studies have described either down- or upregulation of several other peptides, such as CGRP, VIP and NPY, after this type of lesion. The peptide galanin is present at very low levels under normal circumstances, but is seen after axotomy in more than half of all ganglion neurons. With regard to functional significance galanin may act as an endogenous analgesic compound, similar to the opioid peptides, although galanin is present in sensory neurons. (The basal ganlia) Neuropeptides occur abundantly in the basal ganglia, especially in the ventral mesencephalon and the striatum. There is now strong evidence that both substance P, dynorphin as well as enkephalin neurons in the striatum are regulated by the dopaminergic input. Recently we have analysed neurotensin (NT) in the striatum and substantia nigra. After treatment with met-amphetamine there is a marked increase in NT-like immunoreactivity in fibers in the zona reticulata of the substantia
