H_2O_2 production in opsonized zymosan stimulated Suncus murinus macrophage and polymorphonuclear leukocyte, and its differences in animal species

H_2O_2 production stimulated by opsonized zymosan particles (OPZ) was electron cytochemically demonstrated in the resident peritoneal and alveolar macrophages (PM and AM), and polymorphonuclear leukocyte (PMN) from several animals. Adults conventional male animals were used. Resident PM washed out from the peritoneal cavity, and AM were prepared by tracheobronchial lavages. Peripheral blood was harvested from the same animals, and PMN were isolated by a Ficoll-Hypaque method. Then, all the cells were incubated in a medium containing CeCl_3, NaN_3, sucroce and tris-maleate buffer. Following the cytochemical reaction, the cells were fixed in 2% glutaraldehyde. Ultrathin sections were examined under electron microscopy without uranyl or lead staining. PM, AM and PMN showed a similar OPZ-phagocytosis among animals. The electron-opaque cerous precipitate that marked the site of H_2O_2 production was observed on the plasma membrane only at area in contact with OPZ and the intensity decreased in the followed order: Luncus murinus, hamster and rat in PM, AM and PMN. These results suggest that OPZ-stimulated free radical production is varied among animals and that the PM, AM and PMN of Suncus murinus positively used it.
