PCNA/cyclin Immunohistochemistry on testicular atrophy induced by a plasticizer

Histological changes of rat testes induced by the administration of a plasticizer were examined in order to throw light on the mechanism of testicular atrophy. Materials and Methods:Male Wistar rats were fed a diet containing DEHP, a plasticizer, for 2, 4, 8 weeks after birth. For the control experiments, they were fed normal diet. Testicular tissued were fixed in paraformaldehyde or methacarn solutions and embedded in paraffin. Their sections were stained using a monoclonal antibody for PCNA/cyclin by ABC method. Results: Marked decrease in the weight of testis and atrophy of seminiferous tubules were observed in the tissues of four and eight week-old rats, in contrast, the change was not evident in testicular tissues of two weeks-old rats. By immunohistochemical staining for PCNA/cyclin, spermatogonia were positive in normal control tissues. The number of cells positive for PCNA/cyclin decreased in the tissues of rats fed DEHP for four weeks and immuno-positive cells were absent in the tissues of rats fed DEHP for eight weeks. No evident change in the number of immuno-positive cells were observed in two week-old rats. Sertori cells were present in the seminiferous tubules in every experimental group. Conclusion: PCNA/cyclin a maker for S-phase cells, was shown to be useful in immunohistochemical detection of spermatogonia in rat seminiferous tubules. Testicular atrophy was shown to be a result of disappearance of germinal epithelial cells.
