IMMUNOFLUORESCENT STUDIES ON TYROSINE HYDROXYLASE : APPLICATION FOR ITS AXOPLASMIC TRANSPORT

Specific antiserum to bovine adrenal tyrosine hydroxylase (TH) was produced in rabbits, and applied to the immunofluorescent study of the enzyme. Specific immunofluorescence for the enzyme was observed in guinea pig and bovine adrenal medulla and locus coeruleus of rats. Immunofluorescent pictures of TH accumulated in the sciatic nerves of rats and guinea pigs after ligation were first shown in this study. The morphological localizations of TH were compared with biochemical ones and both localizations were in close agreement. Retrograde axoplasmic transport of TH was shown by immunofluorescence on the first day after ligation. The comparison between TH and dopamine β-hydroxylase (DBH) was also made from the point of view of their localizations in vivo by immunofluorescence.
