Immunological Response of Mononuclear Cells from Peripheral Blood and Regional Lymph Nodes in Lung Cancer Patients with Combination of OK-432 and rIL-2

The immunoenhancing effect of a combination of OK-432 and rIL-2 was analyzed using peripheral bloodmononuclear cells(PBMC) and regional lymph node mononuclear cells (RLNMC) of lung cancer patients. With respect to mononuclear cell subsets, increases in CD3, CD4, HLA-DR, and CD25 were observed in PBMC in response to the addition of OK-432 and / or rIL-2, but the increases in CD3 and CD4 were observed in the complete medium alone. In the case of RLNMC, no changes were noted other than increases in HLA-DR and CD25. Moreover, since killer activity disappeared when HLA-DR positive cells were eliminated, it appears that HLA-DR and / or CD25 positive cells are very important in the enhancement of killer activity by OK-432 and / or rIL-2. With regard to cell proliferation capacity, marked proliferation effects were observed in response to interaction between OK-432 and rIL-2 when OK-432 was added first(OK-432→rIL-2) and when OK-432 and rIL-2 were added simultaneously. In the case of killer activity, on the other hand, no difference was detected between the addition of rIL-2 alone and the addition of the combination of OK-432 and rIL-2 to PBMC, however, in the case of RLNMC, higher killer activity was observed in response to OK-432→rIL-2. Thus, it seems that OK-432→rIL-2 is useful in inducing killer cells.
