Studies were undertaken for isolation and culture of atypical mycobacteria from dental calculus of out-patients who were suffering from soft tissue diseases such as gingivitis and alveolar pyorrhea though otherwise healthy. Results are summarized as follows. 1. A total of 38 strains (3.6%) could be detected from a total of 1030 cases of dental calculus, and of these 10 strains (0.97%) could be recovered as pure culture. 2. When classified in Runyon method, these cultures consisted of 6 strains of scotochromogen (60%) and 4 strains of nonphtochromogen (40%) No strain of photochromogen and rapid grower could be isolated. 3. Quantitative organ culture and pathohistologic examination were conducted on mice infected with these organisms, and two strains referred to as Number 1 and Number 7 were found pathogenic to the animal. 4. All the Strains grew colonies within period of 1 week under the temperature of 20 ℃, indicating that they were of quite distinct origin as compared to M. homin isand belonged to saprophytic mycobacteria. 5. When classified according to the color of colonies, there were 4 strains (40%) of light white, 4 strains (40%) of yellow and 2 strains of orange (20%). 6. Biochemical tests for niacin, nicotinamidase, urease and formamidase conducted on these strains also pointed to the saprophytic mycobacteria, and this was further corroborated by the total negative response of these organism to the niacin test and total positive response to the formamidase test. 7. Two scotochromogens of orange color exhibited biological and drug-sensitive characteristics almost similar to those of Ishii strain, but differed from the latter with regard to amidase test. This would be futher evidence that they were saprophytic scotochromogens. 8. Drug sensitivity tests showed that only Number 4 strain exhibited sensitivity to streptomycin and kanamycin at the dose below 1 gamma, and remaining 9 strains were sensitive to streptomycin at 100 gamma and to INAH at 10 gamma. All strains were more or less sensitive to 100 gamma of PAS and 9 of these were fairly sensitive to 100 gamma of kanamycin. 9. when these organisms were compared with Ishii strain and other sapropytes mycobacteria for their biological characteristics, the most marked differences proved to lie in the growth rate, acid fastness and particularly in heat stability.
