Three-week-old male rats of Wistar strain were given a calcium-deficient diet for three weeks and experimental bone debility was induced in them. The rats were then given a standard diet containing calcium and a physiologic active substance (1α-OH-D_3, Calcitonin) for two weeks. The effect of the therapy on the mandibular condyle at growth stage was investigated. The results were as follows : I. General tissue findings In the calcium-deficient diet group as compared with the standard diet controls, the mandibular condyles showed increase in chrondrocyte layers, decrease in calcified matrix surrounding hypertrophic chondrocytes especially, deformation of vacuole and nucleus, and assembly in part. Trabeculae in subchondrocyte bone formation zone decreased and trabeculae were mostly in the low calcification layers. The calcium-deficient diet + standard diet group I and group II showed about the same findings. As compared with the calciumdeficient diet group, calcified matrix surrounding hypertrophic chondrocytes increased ; findings of erosion by chondroclasts increased ; and subchondrocyte bone formation increased. Low calcification layers were seen in part. In the calcium-deficient diet + standard diet + active type vitamin D_3 + ECT group as compared with the calcium-deficient diet + standard diet + ECT + active type vitamin D_3 group, the mandibular condyles showed decrease in hypertrophic chondrocytes, increase in findings of erosion of calcified matrix by chondroclasts, and increase in trabeculae in subchondrocyte bone formation zone. II. Transmission electron microscopic findings The calcium-deficient diet group as compared with the standard diet controls showed increase in the number of osteoclasts in comparison with that of osteoblasts. Cytoplasm showed enlargement of rough surfaced endoplasmic reticulum, well-developed mitochondria and marked increase of mitochondrial cristae. Ruffled borders next to the bone increased. Large and small vacuoles were seen and fragments of collagen fibrils were contained. The calcium-deficient diet + standard diet group I and group II were characterized by active apperances of osteoblasts and osteoclasts and increase in young oseteocytes, as compared with the calcium-deficient diet group. Circularoid osteoblasts increased and rough surfaced endoplasmic reticulum and mitochondria in the cytoplasm were well developed. At the same time, appearances of osteoclast were also active and ruffled borders adhering to the bone were also active. Accmulation of lysosome around mitochondria and vacuoles was seen sporadically. In the caicium-deficient diet + standard diet + active type vitamin D_3 + ECT group and the calcium-deficient diet + standard diet + ECT + active type vitamin D_3 group, osteoblasts began to show many circularoid cells and organelle were also well developed. In harmony with this, minute collagen fibrils appeared actively and accelerated calcification of bone. Osteoclasts adhered to the bone and formed resorption surface. Clear zone and ruffled border continued and the portion facing ruffled border in particular showed collagen fibrils in loosened form. Smoothing of dendric cristae of ruffled border wascharacteristic. Many findings of gap-junction between osteoblastsand osteocytes with cytoplasmic fine projection extending were observed.
