Peritoneal cells (PEC) were induced in rats, guinea pigs, and mice by injection of several antigens such as, liquid paraffin and Corynebacterium equi (C. equi), Escherichia coli (E. coli), and lipopolysaccharide (LPS). Peritoneal cells were composed mainly of two cell-populations, polymorphonuclear leukocytes (PMN) and macrophages (Mφ). These cell numbers were measured after the injection of these antigens. The results obtained were as follows : 1. After the injection of the liquid paraffin and C. equi, PMN were dominant in number to Mφ in PEC both in rats and in mice. The result was opposite in the case of guinea pigs. In all animals, PMN decreased in number in the course of time after that injection. On the contrary Mφ increased. 2. After the injection of the liquid paraffin and C. equi, phagocytosis was frequently seen in PMN both in rats and in mice and was frequently done in Mφ in guinea pigs. 3. After the injection of the killed E. coli to mice, PMN were obtained in almost the same number as Mφ on the 1st day. The PMN number decreased on the 2nd day and increased again on the 3rd day. On the other hand Mφ increased on the 2nd day and decreased on the 3rd day. 4. After the injection of the killed E. coli to guinea pigs, Mφ was dominant in number to PMN during 3 days. 5. After the injection of LPS to mice, Mφ was dominant in number to PMN upto the 2nd day. On the 3rd day PMN increased in number and Mφ decreased. 6. After the injection of LPS to guinea pigs, PMN was dominant in number to Mφ on the 1st day. Then PMN kept decreasing in number for 2 days and Mφ kept increasing. From these results PMN will respond intensely to any killed bacterial injection and Mφ will respord and weakly. It was presumed that LPS directly activated the complement pathway in this peritoneal system.
