In extension of a previous study on the composition of bovine submaxillary gland mucin, an attempt was made to analyze the amino acid sequence on some of the oligopeptides obtained through a series of time-consuming fractionating procedures. The procedures consisted of a minor modification of that of Tettamanti and Pigman involving extraction of mucin from the gland, cetyltrimethylammonium bromide precipitaticn, separation by hydroxyapatite adsorption, desialization followed by Pronase digestion, column chromatography on Sephadex G-10 and then on Dowex 50 resin and finally of high-voltage paper electrophoresis. The electrophoresis was conducted on that portion of eluates from the resin column which appeared in succession to a large ninhydrin positive fraction and which showed both hexosamine and peptide positive reactions. This portion gave 8 distinct bands of which the fast-moving 3 bands proved homogeneous by the 2nd electrophoresis with different buffer and were served for further study. Results showed that they were consisted of 5 to 6 amino acid residues of which the last 3 residues Arg-Gly-Ala were common to all. Occurrence of such unit repetition of amino acid residue through the glycopeptide backbone of mucin is suggested.
