Three-week-old rats corresponding to the human wean period were used and the relation between high protein diet intake and calcium absorption in membranate ossification in the tibia was investigated for those given high protein diet for three weeks (6 weeks old) [I] and those given high protein diet for six weeks (9 weeks old) [II]. The results were as follows : 1. Radiological bone density findings No significant differences were found between control group I and high protein diet group I. Between control group II and high protein diet group II, significant differences were found in interosseous cortex at 1% significance level and in medial cortex at 5% significance level. control group II showed higher values. 2. Analytical findings on Ca and P on the X-ray microanalyzer In the quantitative analysis of Ca, significant differences were found at 5% significance level between control group II and high protein diet group II, and control group II showed higher values. 3. Histopathological findings In high protein diet group I as compared with control group I, location of Haversian lamellae in the whole osseous layer in medial cortex was found to be wide. In interosseous cortex as compared with that in control group I, outer circumferential lamellae and inner circumferential lamellae were wide and seen clearly. Osteoblasts were arranged regularly on their surfaces. In control group II as compared with control group I, osseous tissue was formed thick on the whole and increase in haversian systems especially was marked. Medial cortex and interosseous cortex both showed increase in thickness, and inner circumferential lamellae in interosseous cortex showed increase in thickness in particular. High protein diet group II as compared with control group II showed no changes in the morphology of osseous tissue, but decrease in Haversian systems was found in the whole osseous layer. Also in medial cortex, decrease in bone lacunae was found. 4. Scanning electron microscope findings In the bone formation surface in control group I, many circle-like and oval shaped bone lacunae were found but a number of them were imperfect and had indistinct borders with the surrounding matrices. Bone resorption surface was found extensively adjacent to bone formation surface, and the cavity bottom was shallow. High protein diet group I like control group I showed many bone lacunae in the bone formation surface and both distinct and indistinct borders with the surrounding matrices were found. High protein diet group II as compared with control group II showed no changes in morphology, but bone lacunae in the bone formation surface were irregular in shape and the borders with the surrounding lacunae were indistinct. Bone resorption surface adjacent to bone formation surface increased as compared with that in control group II. 5. Hematologic findings No changes at all were found in any group in electrolytes in serum or in chemical examinations.
