An enzyme-linked immunosorbent assay (ELISA) utilizing avidin-biotin binding was developed for measuring antibodies to capsular-like serotype-specific polysaccharide antigens (SPAs) of Actinobacillus (A.) actinomycetemcomitans serotypes a, b and c. The SPAs were conjugated with biotin. Biotinylated SPAs were immobilized on avidin-precoated wells. The wells immobilized with SPAs reacted specifically with rabbit antisera against whole cells of the corresponding serotype of A. actinomycetemcomitans. This assay will detect sensitively antibodies to SPAs of A. actinomycetemcomitans in sera of periodontitis patients. To investigate the functional activity of antibodies to serotype-b specific antigen (SPAb) of A. actinomycetemcomitans, BALB/c mice were immunized subcutaneously or intranasally with bovine serum albumin-conjugated SPA (SPAb-BSA). Serum IgG responses to SPAb were induced by both subcutaneous and intranasal immunization. Opsonic activity of antisera to SPAb from BALB/c mice was assesed by chemiluminescence assay and phagocytic killing assay using human polymorphonuclear leukocyte. Murine antisera to SPAb efficiently opsonized A. actinomycetemcomitans serotype b in the presence, suggesting that antibodies to SPAb in sera of periodontitis patients may protect the host from infection with the organism.
