PGE_2 and IL-1 were the most important candidates for inducers of bone resorption accompanied by periodontitis. The increase in bone resorption is caused by osteoclast formation and by osteoclastic activation. To assess the contribution of osteoclast formation to the increase in the bone resorption in periodontitis, the effect of PGE_2 and IL-1 on osteoclast formation was examined. Osteoclast formation was evaluated by the appearance of tartarate resistant acid phosphatase (TRAP)-positive cells in mouse bone marrow cells of one-week-culture and two-week-culture. The results were follows : 1. In the culture added 10^<-6>M of PGE_2 many TRAP-positive cells were detected at the first week. Most of these cells disappeared at the second week. The multinucleated TRAP-positive cells were also detected at the first week, and the number decreased thereafter. The propotion of multinucleated TRAP-positive cells to total TRAP-positive cells was about 10%. The areas of resorption pits were observed in the surface of bone slices on which the bone marrow cells were incubated for a week. The areas of the pits increased incubated for two weeks. 2. In the culture added 100U of IL-1 a few mononucleated TRAP-positive cells were detected at the first week, and these cells disappeared thereafter. No multinucleated TRAP-positive cells were detected at the first or the second week. No resorption pits were detected at any week 3. The PGE_2 in the range from 10^<-8>M to 10^<-6>M had dose-dependent increasing effect on the formation of TRAP-positive cells. The PGE_2 higher than 10^<-6>M induced only a small number of TRAP-positive cells. On the formation of multinucleated TRAP-positive cells the PGE_2 in the range from 10^<-8>M to 10^<-5>M had dose-dependent increasing effect. 4. A few TRAP-positive cells were detected in the culture added 100U of IL-1. In the culture added the IL-1 lower than 100U, no TRAP-positive cells were detected. 5. The PGE_2 from 10^<-9>M to 10^<-7>M in combination with 10U of IL-1 enhanced the formation of TRAP-positive cells by nearly 10 times. The IL-1 from 1U to 10U in combination with 10^<-7>M of PGE_2 did not enhance the formation of TRAP-positive cells. The IL-1 of high concentration as 100U in combination with the PGE_2 suppressed the formation. 6. The PGE_2 from 10^<-9>M to 10^<-7>M in combination with 10^<-9>M of activated vitamin D_3, which induced the formation of a few TRAP-positive cells, enhanced the formation of TRAP-positive cells. When the IL-1 from 1U to 10U instead of PGE_2 was added to the culture in the presence of the activated vitamin D_3, the enhancive effect was also recognized. The IL-1 of high concentration as 100U suppressed the formation. These results show as follows : The optimal concentration of PGE_2 for the formation of mononucleated osteoclasts is around 10^<-6>M, and that for the multinucleated cells is over 10^<-5>M. PGE_2 has synergy with IL-1 in the osteoclast formation. Over the optimal dose of these substances suppressed each other. These results suggest the possiblity that PGE_2 has synergistic interact with IL-1 in osteoclast formation to increase the bone resorption in periodontitis.
