Three-week-old Male rats of Wistar strain were given 0.2u/kg of Calcitonin (eel calcitonin) daily and its effect on the mandibular condyle, tibia epiphysis, and tibia metaphysis was examined radiologically and histopathologically. The results were as follows : I. Radiological findings 1) With elapse of experimental days, the peripheral compact osseous layer in the mandibular condyle and in the tibia epiphysis thickened, and trabeculae increased and were running more compactly and about vertically from the articular surface. 2) In the tibia metaphysis, trabeculae increased and were running more compactly and vertically from the surface of the tibia epiphysis with elapse of experimental days. II. Histopathological findings 1) In the mandibular condyle, the cartilage layer thinned owing to decrease in hypertrophic chondrocytes at the third and the fifth week. At the fourth week, no changes in the cartilage layer were found as a result of increase in proliferative chondrocytes and decrease in hypertrophic chondrocytes. Increase in findings of erosion of calcified matrix by chondroclasts and in subchondrocyte bone formation became prominent with elapse of experimental days, and no osteoclasts were observed. 2) In the tibia epiphysis, the cartilage layer thickened because of increase in proliferative chondrocytes and decrease in hypertrophic chondrocytes at the third and the fifth week. At the fourth week, increase in proliferative chondrocytes and decrease in hypertrophic chondrocytes were seen ; however, no changes in the cartilage layer were found. Increase in findings of erosion of calcified matrix by chondroclasts and in subchondrocyte bone formation were noticeable with elapse of experimental days, and no osteoclasts were observed. 3) In the tibia metaphysis, no changes were seen in the cartilage layer at the third week ; however, increase in proliferative chondrocytes and decrease in hypertrophic chondrocytes were marked with elapse of experimental days. Increase in findings of erosion of calcified matrix by chondroclasts and in intracartilage bone formation were conspicuous with elapse of experimental days, and no osteoclasts were observed. 4) In the mandibular condyle under the scanning electron microscope, chondrocyte lacunae in the hypertrophic zone decreased, and calcospherite structures and collagen fiber networks were observed on the floors of the lacunae. Bone lacunae were found in the bone formation zone, and openings of canaliculi were seen on the floors. Collagen fiber networks were also seen. On the basis of the foregoing findings, administration of Calcitonin facilitated cartilaginous tissue growth and bone formation and inhibited osteoclasts in the mandibular condyle, tibia epiphysis, and tibia metaphysis. The effect of Calcitonin was the weakest in the mandibular condyle with the tibia epiphysis and tibia metaphysis following.
