Extracellular glucans produced by Streptococcus mutans contains α-1, 6 and α-1, 3 glucosidic linkages and are closely connected in the pathogenesis of dental caries. In this study, dextranase (EC 3.2.1.11) was detected in the culture supernatant of S. mutans AHT and partially characterized. 1. The cultivation of S. mutans AHT with a complex medium supplemented with glucose resulted in the appearance of the constitutive dextranase in the culture supernatant. 2. The extracellular dextranase was partially purified from the culture supernatant of S. mutans AHT by the fractionation and Bio-Gel A-5m gel filtration. The dextranase fraction (β-fraction) with GTF action and the GTF fraction (α-fraction) without dextranase were separated by the gel filtration ; A 560-fold purification was obtained for dextranase with a yield of 19% over the culture supernatant and a 75-fold purification for GTF with a yield of 9%. The purified dextranase had a pH optimum of ca.5.5, a temperature optimum of ca.37℃ and did not degrade insoluble glucan derived from S. mutans AHT.
