ENZYMATIC ACTIVITY DETERMINATION OF ALKALINE PHOSPHATASE DISSOLVED IN NATURAL WATERS BY FLUOROMETRY

The sensitive determination of alkaline phosphatase activity using 4-methylumbelliferyl phosphate (4-MUP) as a substrate has been investigated by fluorometry. The detection limit of the present method was 5 pM, and the calibration curve was linear up to 3 μM. The experimental conditions for the fluorometric method was strictly compared with those for the conventional spectrophotometric one examined previously. The former was more sensitive by ca. 400-fold than the latter. The method for alkaline phosphatase activity measurement was applied to the chromatographic assay of alkaline phosphatase dissolved in lake and sea waters.
