The purpose of this study was to develop electrochemical assays of citrate and L-lactate in food and biological samples using enzymatic conversions. First, differential pulse polarographic (DPP) determinations of these species with soluble enzyme preparations have been investigated, and DPP measurements of enzyme activity and Michaelis constant (K_m) have been developed. And these enzymes have been immobilized and used in DPP determinations or flow injection analysis. Second, new types of enzyme sensors using a mercury film electrode or differential pulse amperometric detection have been developed. These assays have excellent sensitivity, selectivity, simplicity and rapidity for determinations using small amounts of sample. They were applied to the determinations of citrate and L-lactate in food and biological samples with satisfactory results. It is expected that these assays will be useful for biochemical, clinical and food industrial analysis.
