Different methods, presented by previous investigators, were reexamined by the author, using the yeasts, which were available in Japan. The results were all unsatisfactory. Success was first obtained, when the mixed solution of MgCl_2,MnCl_2,thiamine phosphate buffer and blood samples were directly added to the centrifuged residue of brewers' yeast, which was washed principally after the procedure of Lohmann and Schuster. The production of CO_2 was estimated in usual way. The mean of co-carboxylase content in blood of healthy human beings was found to be 9.7 (and 95% confidence limits were 9.7±0.7μg) per 100ml.
