Assay methods for the fractional estimation of vitamin B_6 components were studied and an improved method was presented. The chief improvements are the following : For estimating pyridoxine, deproteinization is not necessary and for estimating pyridoxamine, urea is dispensable. For zeolite adsorption of pyridoxine the absence of metaphosphoric acid is preferable. Washing with hot water causes the loss of pyridoxine. For extracting pyridoxine and pyridoxamine, preliminary extraction at pH 4.5,and 80° for 15 minutes prior to the hydrolysis in 0.6NH_2SO_4 at 130° for one hour is the optimum condition both for animal and vegetable tissues. The removal of the majority of SO_4 ion by barium favours the recovery of pyridoxine and pyridoxamine after zeolite adsorption. For extracting pyridoxal, direct heating of homogenate in 0.1 N H_2SO_4 at 130° for one hour is the optimum condition. In the blank test heated sample in an acid solution is used as in the main test, followed by the conversion of lactone into 4-pyridoxic acid by heating in an alkaline medium. The content of vitamin B_6 in many animal and vegetable foodstuffs agree quite well with those estimated by animal experiment and microbioassay, which have hitherto been reported in the litrature. The content of 3 forms of vitamin B_6 as well as 4-pyridoxic acid in the blood, and urine was reported. The value of each component of vitamin B_6 in the urine remains the same both with and without acid hydrolysis, indicating that the vitamin occurs in the free form in the urine.
