The kidneys of rats treated for 30 to 50 days with the experimental Vitamin B_6 deficient diet with normal or low magnesium content were examined by electron microscopy. Slight increased number of vacuoles and lysosomal bodies were observed in the proximal convoluted tubuli of vitamin B_6-deficient rats, and the cell debris was prominent in tubular lumen. On the other hand, in the proximal convoluted tubuli of the rats lacking vitamin B_6 and magnesium, the abundant large lysosomal bodies appeared in cytoplasms and the tubular lumen contained the numerous cell debris which had the similar appearance of cellular organelles seen in the proximal tubuli. These observations suggest that the primary step of urinary stone formation is the apatite which is formed in the proximal convoluted tubuli by magnesium depletion and in secondary step the oxalate which is excreted increasingly by vitamin B_6-deficiency precipitates on this apatite.
