Dihydrothiamine (DHB_1) has shown a weak thiamine activity by its oral administration to rats. DHB_1 is stable in the alkaline medium or the artificial intestinal juice ; no thiamine is formed from DHB_1. In the acidic solution DHB_1 is rather unstable and easily oxidized into thiamine at the presence of oxygen. A similar reaction occurred on incubating DHB_1 with the artificial gastric juice, and in the content of rat stomachs or homogenate of them. Both of pepsin and hydrochloric acid, or each of them was effective to convert DHB_1 into thiamine at the presence of oxygen. Even boiled pepsin and protein such as casein, egg albumin or milk stimulated formation of thiamine from DHB_1. It will be reasonably assumed that DHB_1 is partly transformed into thiamine by the contact to acidic gastric juice, thus showing thiamine activity only per os, not by subcutaneous injection.
